6-Hydroxy-1-Naphthoic Acid

CAS2437-17-4

6-Hydroxy-1-naphthoic acid (CAS 2437-17-4), systematically 6-hydroxynaphthalene-1-carboxylic acid, is a naphthalene building block that carries a phenolic hydroxyl at C-6 and a carboxylic acid at C-1. The two groups can be modified separately, which makes it a starting material for 6-alkoxy-1-naphthoic acids, naphthamides and other pharmaceutical intermediates. KingPharm supplies 6-hydroxy-1-naphthoic acid for research and production with a batch-specific certificate of analysis.

Product Description

6-Hydroxy-1-naphthoic acid (C11H8O3, MW 188.18) is a 1,6-disubstituted naphthalene with a carboxylic acid at the 1-position and a phenolic hydroxyl at the 6-position. It is a solid, white to brown in appearance, with a melting point of about 207–212 °C, and is soluble in methanol.

The phenol can be O-alkylated or acylated, and the carboxylic acid can be converted into esters, amides or acid chlorides. This lets chemists build on either side of the naphthalene core. A typical route is O-alkylation to 6-alkoxy-1-naphthoic acids and esters, which have been described as intermediates for aldose reductase inhibitors. Amide coupling at the acid gives naphthamide derivatives.

Compared with simple naphthoic acids, the extra hydroxyl adds a second, chemically distinct point of attachment without further ring functionalisation. Starting from the hydroxy acid also avoids introducing the 6-oxygen substituent at a late stage, where naphthalene regioselectivity is harder to control.

When sourcing, confirm the substitution pattern. 6-Hydroxy-1-naphthoic acid is easily confused with its isomer 6-hydroxy-2-naphthoic acid (CAS 16712-64-4), so check CAS number, NMR and HPLC purity on the certificate of analysis. Isomeric hydroxynaphthoic acids and residual naphthol are the main impurities to review.

Applications

With separate phenol and carboxylic acid handles, 6-hydroxy-1-naphthoic acid is used mainly as a synthetic intermediate.

6-Alkoxy-1-naphthoic acid intermediates

O-Alkylation of the phenol gives 6-alkoxy-1-naphthoic acids and esters, described as intermediates for aldose reductase inhibitors.

Naphthamide derivatives

Amide coupling at the carboxylic acid gives naphthamides, a scaffold used in reported kinase and HDAC inhibitor programmes.

Medicinal chemistry building block

The two independent handles allow stepwise derivatisation of the naphthalene core for structure–activity studies.

Frequently Asked Questions

Is 6-hydroxy-1-naphthoic acid interchangeable with 6-hydroxy-2-naphthoic acid?

No. They are positional isomers (CAS 2437-17-4 vs 16712-64-4) and lead to different products. Check the CAS number and NMR data on the COA before use.

How can only the hydroxyl group be alkylated?

Because the carboxylic acid can also react, a common approach is to esterify the acid first or alkylate both groups, then hydrolyse the ester back to the free acid.

How should it be stored?

Keep the container tightly closed in a cool, dry place, protected from light and away from strong oxidisers. Follow the storage conditions on the SDS.

What is the CAS number of 6-Hydroxy-1-Naphthoic Acid?

The CAS Registry Number of 6-Hydroxy-1-Naphthoic Acid is 2437-17-4. 6-Hydroxy-1-Naphthoic Acid is also known as 6-HYDROXY-1-NAPHTHOIC ACID; OHNA; 6-hydroxynaphthalene-1-carboxylic acid.
